Transcriptomic Analysis for the Identification of Metabolic Pathway Genes Related to Toluene Respons

Toluene is an industrial raw material and solvent that can be found abundantly in our daily life products. The amount of toluene vapor is one of the most important measurements for evaluating air quality. The evaluation of toluene scavenging ability of different plants has been reported, but the mechanism of plant response to toluene is only partially understood. In this study, we performed RNA sequencing (RNA-seq) analysis to detect differential gene expression in toluenetreated and untreated leaves of Ardisia pusilla. A total of 88,444 unigenes were identified by RNA-seq analysis, of which 49,623 were successfully annotated and 4101 were differentially expressed. Gene ontology analysis revealed several subcategories of genes related to toluene response, including cell part, cellular process, organelle, and metabolic processes. We mapped the main metabolic pathways of genes related to toluene response and found that the differentially expressed genes were mainly involved in glycolysis/gluconeogenesis, starch and sucrose metabolism, glycerophospholipid metabolism, carotenoid biosynthesis, phenylpropanoid biosynthesis, and flavonoid biosynthesis. In addition, 53 transcription factors belonging to 13 transcription factor families were identified. We verified 10 differentially expressed genes related to metabolic pathways using quantitative real-time PCR and found that the results of RNA-seq were positively correlated with them, indicating that the transcriptome data were reliable. This study provides insights into the metabolic pathways involved in toluene response in plants.

Saussurea albifolia M. J. Nam & H. T. Im (Compositae), a new species from the Baekdudaegan Area, Kor

Saussurea albifolia, a new species, is reported from the Baekdudaegan Mountains. It has distinctive morphological characteristics that distinguish it from other congeneric species of Saussurea in Korea; radical leaves persist or withered till flowering, white tomentose leaf beneath, campanulate involucre with browncobwebby hairs, and dark purplish phyllaries with acuminate tips. Saussurea albifolia is a new, endemic species, narrowly restricted to the Korean Peninsula as a consequence of adaptation to alpine or subalpine environments of the Baekdudaegan Area. Among the Korean species of Saussurea, S. gracilis Maxim., S. insularis Kitam., and S. seoulensis Nakai are morphologically similar to S. albifolia by having leaves with white hairs beneath and persistent radical leaves during blooming period. It has been well understood that Saussurea is one of the highly diversified and adaptable groups in Asteraceae and also that the currently recognized species in Korea likely significant underestimates its diversity on the Korean Peninsula

OsATL38 mediates mono-ubiquitination of the 14-3-3 protein OsGF14d and negatively regulates the cold

ABSTRACT One of the major regulatory pathways that permits plants to convert an external stimulus into an internal cellular response within a short period of time is the ubiquitination pathway. In this study, OsATL38 was identified as a low temperature-induced gene that encodes a rice homolog of Arabidopsis Tóxicos en Levadura RING-type E3 ubiquitin (Ub) ligase, which was predominantly localized to the plasma membrane. OsATL38-overexpressing transgenic rice plants exhibited decreased tolerance to cold stress as compared with wild-type rice plants. In contrast, RNAimediated OsATL38 knockdown transgenic progeny exhibited markedly increased tolerance to cold stress relative to that of wild-type plants, which indicated a negative role of OsATL38 in response to cold stress. Yeast two-hybrid, in vitro pull-down, and co-immunoprecipitation assays revealed that OsATL38 physically interacted with OsGF14d, a rice 14-3-3 protein. An in vivo target ubiquitination assay indicated that OsGF14d was mono-ubiquitinated by OsATL38. osgf14d knockout mutant plants were more sensitive to cold stress than wild-type rice plants, indicating that OsGF14d is a positive factor in the response to cold stress. These results provide evidence that the RING E3 Ub ligase OsATL38 negatively regulates the cold stress response in rice via mono-ubiquitination of OsGF14d 14-3-3 protein.

Sisymbrium irio L. (Brassicaceae): a new alien plant in Korea

ABSTRACT As anthropogenic transports of biomaterials have rapidly escalated in the past century, biological invasions have been of great concern on a global scale. Given the exponential growth of alien species passing through the initial point of introduction, reporting an alien species on its initial appearance is important. Sisymbrium irio, an annual herb distributed widely throughout the northern hemisphere, is a notorious weed for its strong invasiveness. In 2016 S. irio was first reported in Korea. The plant was found along the urban expressway, Beonyeong-ro, connecting Busan trading port to Gyeongbu expressway. To properly identify the species, we collected ten S. irio samples from the reported site and examine their morphological characters. The identification key of Sisymbrium irio and its related species in the genus was constructed to clarify taxonomic confusions. We further determined the identity of the taxa by inferring the phylogeny of S. irio and the related taxa. To investigate the distribution changes for the past four years since the first observation in 2016, we monitored the area in late summer of 2020. Both morphological and molecular examinations indicated that the Sisymbrium species we found in Korea for the first time was S. irio. From our site monitoring, we newly found a second distribution site that is approximately 1.5 km away from the original site. Considering the geographical proximity of the introduction sites from the port, we hypothesize that S. irio might started expanding its range by the aid of roads. Given the role of roads as a corridor for alien species, further expansion of S. irio in Korea is expected.

Comparative Floral and Pollen Micromorphology of Leonurus japonicus and L. macranthus (Lamiaceae)

Abstract Floral micromorphology and pollen morphology of two Leonurus (Lamiaceae) species were examined and compared using scanning electron microscopy to evaluate the diagnostic value of these features to facilitate future studies on floral biology and taxonomy. Floral epidermal patterns were similar in both species, with the conical to central-conical epidermal cells on the adaxial side of the lower lip (corolla). Capitate, peltate, scale-like glandular, and non-glandular trichomes were distributed on the surface of the floral organs. Notably, scale-like anther glands and floral stomata were found on the anthers and abaxial side of the calyx, respectively. Pollen grains had bi-reticulate exine with angular primary lumina and rounded secondary lumina. These characteristics provide indirect evidence of a close association between plant-pollinator interactions and effective pollination. In addition, quantitative traits of pollen grains and trichome types on the adaxial side of the lip differed between the two species. These characteristics may have diagnostic and taxonomic value for the genus Leonurus and family Lamiaceae.

Non-Deep Simple Morphophysiological Dormancy and Germination Characteristics of Gentiana triflora va

ABSTRACT This study investigated the kind of seed dormancy and seed germination of Gentiana triflora var. japonica (Kusn.) H. Hara for developing a seed propagation method. The seeds were collected in October 2020 from plants at Mt. Sobaeksan, Korea. In a water imbibition experiment, seed weights increased by >101.9% of their initial masses over 12 h. Effects of incubation temperature (5, 15, 20, 25, 15/6, or 25/15 C), cold stratification period (5 C; 0, 4, 8, or 12 weeks), and gibberellic acid (GA3; 0, 10, 100, or 1000 mgL?1) and potassium nitrate treatment (KNO3; 0, 1000, 2000, or 4000 mgL?1) on seed germination were investigated to characterize seed dormancy. These seeds exhibited underdeveloped embryos during seed dispersal. The seeds failed to reach the final germination of 15.0% after treatment at 5, 15, 20, 25, 15/6, or 25/15 C. After cold stratification for 8 weeks, the germination increased dramatically by >90.0% compared to that at 0 weeks. After the GA3 treatment, the germination reached >80.0% within 5 days. The final germination was 90.0% in the 100 mgL?1 GA3 treatment group. However, the KNO3 treatment had no effect on seed germination. Therefore, the G. triflora var. japonica seeds exhibited non-deep simple morphophysiological dormancy.

백두대간 지리산권역(육십령-천왕봉 구간) 마루금의 식물사회학적 유형분류 및 군집 특성

요 약: 본 연구는 국토의 생태축이라고 할 수 있는 백두대간의 지속가능한 보전과 관리방안 수립에 기여하고자 실시되었으며, 식물사회학적 유형분류를 통해 종조성을 파악하고 식생단위별 군집 특성을 파악하였다. 백두대간(육십령-천왕봉 구간) 마루금 주변을 대상으로 2020년 5월부터 9월까지 총 373개소의 산림식생조사를 실시하였다. 식생유형분류 결과, 9개의 식생단위로 구분되었으며 정성적 식생유형분류를 정량적 DCA 분석을 통해 검증하였다. 중요치 분석 결과, 아고산지대에 자생하는 종조성 으로 묶인 식생단위 1∼5는 구상나무, 가문비나무, 잣나무, 사스래나무 등 아고산식생 위주로 중요치가 높게 나타났으며, 해발고도가 높지 않은 마루금에서는 능선 위주의 지형 특성상 소나무, 졸참나무 등의 중요치가 높게 나타났다. 종다양도 분석 결과, 평균해발고도가 상대적으로 높은 구상나무군락(식생단위 1∼5)이 다른 식생단위에 비하여 상대적으로 종다양도가 높게 나타났다. NMS 결과 및 환경인자와의 상관분석 결과, 식생단위 1, 2, 4, 5와 해발고도와의 양의 상관관계를 가지는 것으로 나타났다. 본 연구는 소나무군락, 비목군락의 저해발 지역 식생부터 구상나무군락, 가문비나무군과 같은 고해발 지역 식생까지 모두 포함되었고 습윤한 지역의 계곡부 식생이 포함되지 않은 백두대간의 능선 식생을 특성을 나타내었다.

A New Lichen-Forming Fungus, Aspicilia humida, from a Forested Wetland in South Korea

Abstract: Aspicilia humida Lee is described as a new lichen-forming fungus from a wetland forest, South Korea. The new species is distinguishable from Aspicilia aquatica (Fr.) Korb., the most € similar species, by the absence of prothallus, black disk without green color in water, olivebrown epihymenium, shorter hymenium, hymenium I þ yellowish blue-green, wider paraphysial tips without a vivid pigment, smaller asci, smaller ascospores, and the presence of stictic acid. Molecular analyses employing internal transcribed spacer (ITS) and mitochondrial small subunit (mtSSU) sequences strongly support A. humida as a distinct species in the A. cinerea group. A surrogate key is provided to assist in the identification of all 28 aspicilioid species of Korea.

A New Lichenized Fungus, Psoroglaena humidosilvae, from a Forested Wetland of Korea

Abstract: Psoroglaena humidosilvae Lee is described as a new lichen species from a wetland forest in South Korea. The new species is distinct from P. stigonemoides (Orange) Henssen by little projections locally present on the thallus; smaller, paler, and globose perithecia; smaller asci; and smaller ascospores generally 3-septate. Molecular analyses employing internal transcribed spacer (ITS), mitochondrial small subunit (mtSSU), and nuclear large subunit ribosomal RNA (LSU) sequences strongly support P. humidosilvae as a nonidentical species in the genus Psoroglaena. A surrogate key is provided to assist in the identification of all 22 species of Psoroglaena.

Exploring the Outdoor Recreational Behavior and New Environmental Paradigm among Urban Forest Visito

ABTRACT This is international comparative research on the perception of local residents toward the natural environment in South Korea, Taiwan, and Indonesia. Through the New Ecological Paradigm (NEP) investigation, perceptions of natural environmental conservation and utilization of 664 urban forest visitors were analyzed, and the relationship between recreational behavior, NEP scores, and demographic characteristics was investigated. The three countries, with different histories, cultures, and economic development, showed statistically significant differences in all items. In terms of the NEP response score, Taiwan showed the most positive results with an average of 4.08. Frequent visits by the elderly and family were common significant factors of high NEP score for all survey locations. In the atory factor analysis of latent variables for NEP, 'limits to growth' were significant in South Korea while 'ecological crisis' was more significant in Taiwan and Indonesia.
