Comparison of species composition among Picea jezoensis (Siebold & Zucc.) carrière forests

Abstract: In this study, we aimed to provide basic data on the vegetation structure of the Picea jezoensis forest of Gyebangsan, Sobaeksan, Deogyusan, and Jirisan in South Korea and Baekdusan in China and to compare the abiotic and biotic factors correlated with the species composition of Picea jezoensis forests from South Korea and China. The Picea jezoensis forest stand structure was more stable in the Baekdusan region of China than in South Korea. In Sobaeksan, South Korea, a severe decline in Picea jezoensis was observed, and the basal area occupied by the species (2.89 1.11 ㎡/ha; approximately 8% of the total basal area) was lower than that occupied by broad-leaved trees.

Abstract : Alien invasive species are introduced with or without intent and spreading all over Korea. They are known to have negative effects on biodiversity such as economic and environmental damage and causing decrease or loss of native species. The habitats like wetland, reservoir and riverside are especially in danger of being invaded by alien species due to stress and disturbance. Therefore, Korea National Arboretum is steadily working on research and studies on managing alien invasive species. This research aims to collect basic information of Ludwigia peploides subsp. montevidensis (Spreng.) P.H. Raven which was found near riverside in Suwon-si and is concerned to become an invasive alien species. We expect the description, diagram and pictures of this taxon will be helpful for early detection and effective management.

A survey of viruses and viroids in astringent persimmon (Diospyros kaki Thunb.) and the development

Abstract: Astringent persimmon (Diospyros kaki Thunb.) is an important fruit crop in Korea; it possesses significant medicinal potential. However, knowledge regarding the pathogens affecting this crop, particularly, viruses and viroids, is limited. In the present study, reverse transcriptionpolymerase chain reaction (RT-PCR) and high-throughput transcriptome sequencing (HTS) were used to investigate the viruses and viroids infecting astringent persimmons cultivated in Korea. A one-step multiplex RT-PCR (mRTPCR) method for the simultaneous detection of the pathogens was developed by designing species-specific primers and selecting the primer pairs via combination and detection limit testing. Seven of the sixteen cultivars tested were found to be infection-free. The RT-PCR and HTS analyses identified two viruses and one viroid in the infected samples (n = 51/100 samples collected from 16 cultivars). The incidence of single infections (n = 39/51) was higher than that of mixed infections (n = 12/51); the infection rate of the Persimmon cryptic virus was the highest (n = 31/39). Comparison of the monoplex and mRT-PCR results using randomly selected samples ed the efficiency of mRT-PCR for the identification of pathogens. Collectively, the present study provides useful resources for developing disease-free seedlings; further, the developed mRT-PCR method can be extended to investigate pathogens in other woody plants.

Morphological and molecular data support the separate species status of Syringa fauriei in Korea

Abstract: Syringa fauriei is an unresolved lilac taxon (Oleaceae) native to specific geographical areas and present in the rivers and valleys of Gangwon province in South Korea. It was first identified on Mt Geumgang by Faurie in 1906 and recognized as an independent species. Nevertheless, S. fauriei has been controversial as a synonymous species for S. reticulata subsp. amurensis without clear evidence. Therefore, we conducted this study on the taxonomic position of S. fauriei by examining the morphological and molecular differences between S. fauriei and its related taxa, the S. reticulata complex. Phylogenetic relationships were also inferred using molecular markers of nuclear ribosomal DNA internal transcribed spacer (ITS1-5.8S-ITS2; ITS) and external transcribed spacer regions. Consequently, S. fauriei and its related taxa were classified into two distinct clades: S. fauriei and the S. reticulata complex. Morphological and molecular data revealed that S. fauriei was endemic to Korea. The species characteristics of S. fauriei were further described for taxonomic re-establishment.

Discrimination of Transgenic Canola (Brassica napus L.) and their Hybrids

Abstract: In recent years, the rapid development of genetically modified (GM) technology has raised concerns about the safety of GM crops and foods for human health and the ecological environment. Gene flow from GM crops to other crops, especially in the Brassicaceae family, might pose a threat to the environment due to their weediness. Hence, finding reliable, quick, and low-cost methods to detect and monitor the presence of GM crops and crop products is important. In this study, we used visible near-infrared (Vis-NIR) spectroscopy for the effective discrimination of GM and non-GM Brassica napus, B. rapa, and F1 hybrids (B. rapa X GM B. napus). Initially, Vis-NIR spectra were collected from the plants, and the spectra were preprocessed. A combination of different preprocessing methods (four methods) and various modeling approaches (eight methods) was used for effective discrimination. Among the different combinations, the Savitzky-Golay and Support Vector Machine combination was found to be an optimal model in the discrimination of GM, non-GM, and hybrid plants with the highest accuracy rate (100%). The use of a Convolutional Neural Network with Normalization resulted in 98.9%. The same higher accuracy was found in the use of Gradient Boosted Trees and Fast Large Margin approaches. Later, phenolic acid concentration among the different plants was assessed using GC-MS analysis. Partial least squares regression analysis of Vis-NIR spectra and biochemical characteristics showed significant correlations in their respective changes. The results showed that handheld Vis-NIR spectroscopy combined with chemometric analyses could be used for the effective discrimination of GM and non-GM B. napus, B. rapa, and F1 hybrids. Biochemical composition analysis can also be combined with the Vis-NIR spectra for efficient discrimination.

Biogenesis and Lipase-Mediated Mobilization of Lipid Droplets in Plants

Abstract: Cytosolic lipid droplets (LDs) derived from the endoplasmic reticulum (ER) mainly contain neutral lipids, such as triacylglycerols (TAGs) and sterol esters, which are considered energy reserves. The metabolic pathways associated with LDs in eukaryotic species are involved in diverse cellular functions. TAG synthesis in plants is mediated by the sequential involvement of two subcellular organelles, i.e., plastids - plant-specific organelles, which serve as the site of lipid synthesis, and the ER. TAGs and sterol esters synthesized in the ER are sequestered to form LDs through the cooperative action of several proteins, such as SEIPINs, LD-associated proteins, LDAP-interacting proteins, and plant-specific proteins such as oleosins. The integrity and stability of LDs are highly dependent on oleosins, especially in the seeds, and oleosin degradation is critical for efficient mobilization of the TAGs of plant LDs. As the TAGs mobilize in LDs during germination and post-germinative growth, a plant-specific lipase—sugar-dependent 1 (SDP1)—plays a major role, through the inter-organellar communication between the ER and peroxisomes. In this review, we briefly recapitulate the different processes involved in the biogenesis and degradation of plant LDs, followed by a discussion of future perspectives in this field.

한국의 묵논에서 멸종위기식물의 복원생태학적 연구 및 서식지 관리방안

Abstract 묵논을 생태적으로 활용하기 위한 방법의 일환으로 논의 특성을 고려하여 생활형이 다른 멸종위기식물 9종의 현지외보전 대상지로서의 잠재력을 확인하였다. 한국의 전통적인 논의 구성요소인 둠벙을 조성하기 위해 묵논의 일부를 변형하여 수분 환경을 다르게 하였다. 묵논에 각 식물들의 유성생식체(seed, sapling)와 무성생식체를 이식하여 1년간 생존률과 식물계 절학 반응을 관찰하였고 그 후 2년간 모니터링을 실시하였다. 그 결과 묵논에 도입이 가능한 수생식물은 4 분류군 (가시연 꽃, 삼백초, 전주물꼬리풀, 조름나물), 지중식물은 1 분류군 (큰바늘꽃), 반지중식물은 1 분류군 (독미나리) 이었다. 도입시 가시연꽃, 전주물꼬리풀, 조름나물은 둠벙에 도입해야하고, 삼백초, 큰바늘꽃, 독미나리는 논습지에 도입해야한다. 또한, 가시연꽃과 순채의 생육은 초본류에 의해 저해되었고 큰바늘꽃의 생식생장은 섭식자에 의해 저해되었다. 따라서 묵논에 도입한 멸종위기식물의 효율적인 정착을 돕기 위해서는 생육을 저해하는 초본류들의 제거와 섭식자의 관리가 필요하 고, 논둑에 목본 또는 식생 매트를 형성하는 초본류를 식재하여 둑의 붕괴를 방지할 필요가 있다. 특히 묵논을 생태적으로 활용할 때 생물다양성을 높이기 위해서는 둠벙을 조성하여 수분환경을 다양화시키는 것이 효과적이다.

DMZ에 인접한 봉화산 일대의 관속식물 분포 연구

Abstract Mt. Bonghwa reaches a height of 800 m (2,870 ft) and stands on the flank of Mt. Daeam-san, which has an elevation of 1,304 m (4,278 ft). It is important to survey the flora of Bonghwa-san in that it is a way to make out what kind of plant species exist in DMZ due to the geographical characteristics of Bonghwa-san adjacent to Korean DMZ. Therefore this study was conducted to elucidate the distribution of vascular plants and their value of Bonghwa-san. The vascular plants that were collected 10 times from May 2015 to April 2017 consisted of a total of 525 taxa; 93 families, 309genera, 458 species, 4 subspecies, 58 varieties and 5 forms. The observed rare plant species as designated by Korea Forest Service were 12 taxa including Echinosophora koreensis, Epimedium koreanum, Anemone koraiensis etc. Endemic species were 16 taxa containing Cirsium setidens, Heloniopsis koreana, Salvia chanryoenica etc. 31 taxa were recorded as naturalized plants and their naturalized rate and urban index were noted as 5.8% and 7.25%. Among the surveyed 525 taxa, edible, medicinal, pasturing, ornamental, timber, dye, fiber and industrial plants included 213 taxa, 172 taxa, 196 taxa, 50 taxa, 23 taxa, 10 taxa, 6 taxa and 3 taxa, respectively.

천축산 일대 산림습지의 식생다양성과 관리방안

요약 경상북도 울진군 서면 왕피천 유역 생태·경관 보전지역 내 천축산 자락의 고해발지역에 위치한 4개의 소규모 산지습지 식물군락의 다양성과 그 분포 특성을 밝히고자 한다. 서식처 유형과 종조성의 균질성을 고려한 Z.-M. 학파의 식물사회학적 식생조사 방법에 따라 총 26개의 식생자료가 수집되었다. 우점 상관에 따른 식생 구분과 종조성을 고려한 식생 유형분류를 통해 9개 단위식생으로 구성된 4개 상관형의 식생 분포가 확인되었다. 진퍼리새-꽃창포군락은 연구지역을 대표하면서 우점하는 식물군락이다. 인간의 이용 이후 방치된 균질한 입지에 자연천이로 식생이 발달하고 있으나 면적이 좁아 기타 식물군락의 분포는 드물게 관찰되었다. 습지별 미세지형과 수문환경은 식생의 다양성과 분포에 영향을 미치는 핵심요소로 확인되었다.
