Detection, isolation, and characterization of the cucumber mosaic virus in Pseudostellaria heterophy

Abstract Weeds play an important role in the survival of viruses and are potential inoculum sources of viral diseases for crop plants. In this study, specimens of Pseudostellaria heterophylla exhibiting symptoms of the cucumber mosaic virus (CMV) were collected in Bonghwa, Korea. The characteristics of the disease were described and leaf RNA was extracted and sequenced to identify the virus. Three CMV contigs were obtained and PCR was performed using specific primer pairs. RNA from positive samples exhibiting CMV leaf symptoms was amplified to determine the coat protein. A sequence comparison of the coat protein gene from the CMV BH isolate shared the highest nucleotide identity (99.2%) with the CMV ZM isolate. Phylogenetic analysis showed that CMV-BH belonged to subgroup IA and that the most closely-related isolate was CMV-ZM. All test plants used for the biological assay were successfully infected with CMV and exhibited CMV disease symptoms such as blistering, mosaic, and vein yellowing. To our knowledge, this is the first report of CMV infection in P. heterophylla from Korea

파주시 하천변에 서식하는 층층둥굴레의 계절학과 번식생태학적 특성

요 약 층층둥굴레는 백합과에 속하고 한국에 자생하는 다년생 초본이다. 이는 멸종위기생물종 목록에서 해제되었지만 하천변에 서식하기 때문에 빈번한 교란으로 개체군은 훼손 등 위협에 처해있다. 따라서 층층둥굴레의 기초 생태학적 특성을 밝히기 위해, 본 연구는 2년 동안 파주시의 하천변에서 자생하는 층층둥굴레의 지상부의 식물계절학 조사와 번식기관인 꽃과 열매의 수를 측정하였다. 그 결과, 층층둥굴레의 지상부는 4월 말에 출현하고 5월 말까지 약 1m로 빠르게 생장하였다. 5월 초, 1개의 꽃자루 당 2개의 꽃봉오리가 줄기 마디에 최대 6쌍까지 형성되었고, 5월 중순부터 개화하였다. 열매는 6월 중순부터 형성되어 9월 말까지 성숙하였다. 층층둥굴레의 꽃과 열매는 줄기의 기저부부터 정단부까지 차례로 달렸고, 꽃과 열매의 수는 줄기의 중앙 부분에서 가장 많이 형성하였고, 지면과 가장 가까운 마디부터 최대 5마디까지는 형성하지 않았다. 본연구에서 밝힌 하천변에서 자생하는 층층둥굴레의 기초 생태학적 특성은 이들의 보전을 위한 기초 자료로 유용할 것이다.

Two New Species of Laccaria (Agaricales, Basidiomycota) from Korea

ABSTRACT Species of Laccaria (Hydnangiaceae, Agaricales, and Basidiomycota) are well-known ectomycorrhizal symbionts of a broad range of hosts. Laccaria species are characterized by brown, orange, or purple colored basidiocarps, and globose or oblong, echinulate and multinucleate basidiospores. While some Laccaria species are easily identified at the species level using only the morphological characteristics, others are hard to distinguish at the species level due to small differences in morphology. Heretofore, ten Laccaria species have been reported in Korea. While studying the fungal diversity in the National Parks of Korea, two new Laccaria species were discovered. Species identification was done based on molecular analyses (ITS, 28S rDNA, rpb2, and tef1), then were ed by their corresponding morphologies. The two newly discovered Laccaria species are proposed here as Laccaria macrobasidia and Laccaria griseolilacina. The unique morphological characters of L. macrobasidia that distinguish it from its closely related species are orange-brown colored basidiocarp, long basidia and the absence of cheilocystidia. L. griseolilacina is characterized by a light grayish lavender-colored pileus and the absence of cheilocystidia. Two new species are described and illustrated in the present paper.

Keyword Analysis of Arboretums and Botanical Gardens Using Social Big Data

Abstract This study collects social big data used in various fields in the past 9 years and explains the patterns of major keywords of the arboretums and botanical gardens to use as the basic data to establish operational strategies for future arboretums and botanical gardens. A total of 6,245,278 cases of data were collected: 4,250,583 from blogs (68.1%), 1,843,677 from online cafes (29.5%), and 151,018 from knowledge search engine (2.4%). As a result of refining valid data, 1,223,162 cases were selected for analysis. We came up with keywords through big data, and used big data program Textom to derive keywords of arboretums and botanical gardens using text mining analysis. As a result, we identified keywords such as 'travel', 'picnic', 'children', 'festival', 'experience', 'Garden of Morning Calm', 'program', 'recreation forest', 'healing', and 'museum'. As a result of keyword analysis, we found that keywords such as 'healing', 'tree', 'experience', 'garden', and 'Garden of Morning Calm' received high public interest. We conducted word cloud analysis by extracting keywords with high frequency in total 6,245,278 titles on social media. The results showed that arboretums and botanical gardens were perceived as spaces for relaxation and leisure such as 'travel', 'picnic' and 'recreation', and that people had high interest in educational aspects with keywords such as 'experience' and 'field trip'. The demand for rest and leisure space, education, and things to see and enjoy in arboretums and botanical gardens increased than in the past. Therefore, there must be differentiation and specialization strategies such as plant collection strategies, exhibition planning and programs in establishing future operation strategies.

희귀수종 거문도닥나무(Wikstroemia ganpi (Sieb. et Zucc.) Maxim.)의 번식에 관한 연구

Abstract This study was conducted on propagation use for conservation scheme of a threatened plant; Wikstroemia ganpi (Sieb. et Zucc.) Maxim(Geomundo false ohelo). The seed propagation was showed higher (95%) in storage and lower values in ground and cutting. Softwood cutting was higher than hardwood cutting and it was possessing higher ratio of rooting that increase concentration of IBA and NAA. It was determined that for Geomundo false ohelo seedling was effective than cutting. In-situ and ex-situ conservation and restoration of substitute habitats of Geomundo false ohelo is therefore necessary due to human trampling in the habitats, damage, natural selection, loss and suppression.

Plastome comparison and evolution within the tribes of Plantaginaceae: Insights from an Asian gypsyw

ABSTRACT In spite of availability of several plastomes representing different tribes of Plantaginaceae, sparse attempts have been made to understand the plastome structure, evolution, and phylogenomics. In the present study, we have made an effort to understand the gene content and plastome evolution in the family Plantaginaceae using the newly generated plastome sequence of Veronica ovata subsp. kiusiana, a taxon native to SE Asia. In the first-ever attempt, plastomes of seven out of 10 tribes of Plantaginaceae have been compared to understand the evolution across the tribes of lantaginaceae. The size of the plastome of V. ovata subsp. kiusiana is 152,249 bp, showing a typical quadripartite struc- ture containing LSC, SSC, and two IRs with the sizes of 83,187, 17,704, and 25,679 respectively. The plas- tome comparison revealed the unique deletions in ycf2 and ndhF genes of members of different tribes, and also revealed high nucleotide variable hotspots. The study also revealed six highly variable genes and intergenic spacer viz. rps16 , rps15-ycf1, ccsA-ndhD, ndhC-trnV, petN-psbM, and ycf1-trnN as potential DNA barcodes for the genus Veronica . The phylogenomic study revealed the sister relationship between V. ovata subsp. kiusiana and V. persica and also suggested the tentative placement of seven tribes in the fam- ily Plantaginaceae.
