Preventing scattering of Tetranychus urticae in Rosa hybrida through dsCOPB2 expression

Abstract: To obtain rose plants resistant to Tetranychus urticae, which is one of the major pests of roses grown in a plastic house or greenhouse to be harvested as cut-flowers, we introduced the double-stranded RNA of the coatomer protein complex (dsCOPB2) from T. urticae (inserted into the pPZP200-Bar vector) to somatic embryos or embryogenic calluses of a rose breeding line (KR056006), using an Agrobacterium-mediated transformation method. It took 11 to 41 months to obtain eight plants, which were regenerated from the somatic embryos or embryogenic calluses through a selection procedure that uses phosphinothricin acetyltransferase (ppt; 2 mg L − 1 ) after co-cultivation. Validation of the transgene in pseudo-transgenic plants was verified via genomic polymerase chain reaction (PCR) and Southern Blot analyses. It was ed that one to six copies of the transgene were transferred to the eight dsCOPB2-transgenic lines; the levels of the transgene were analyzed via real-time quantitative PCR (RT-qPCR). In the resistance test of one transgenic line (KR056006-COPB2–13) against T. urticae, the pest showed a high mortality rate of approximately (~89%), and egg-laying was relatively decreased by approximately 88%, compared to those for a non-transgenic (NT) rose line, KR056006. This suggests that the transgenic rose plants with high accumulated amounts of dsCOPB2 have a negative effect on the survival of T. urticae. It is expected that these results will accelerate the development of pest-resistant cultivars and may replace conventional pest control methods in general.

A New Lichenized Fungus, Lendemeriella luteoaurantia, with a Key to the Species of Lendemeriella

Abstract : Lendemeriella luteoaurantia B.G. Lee is described as a new lichen species from South Korea. The new species is identified by smaller, yellow-orange apothecia, larger ascospores with wider septum width, and the absence of Cinereorufa-green pigment and teloschistin, different from the closest species, L. aureopruinosa I.V. Frolov, Vondrák, Arup, Konoreva, S. Chesnokov, Yakovczenko and Davydov in morphology and chemistry. Molecular phylogeny employing internal transcribed spacer (nuITS), nuclear large subunit ribosomal RNA (nuLSU), and mitochondrial small subunit (mtSSU) sequences strongly supports the new species as nonidentical in the genus Lendemeriella. A preliminary key is provided to assist in the identification of all 10 species of Lendemeriella.

외부형태학적 형질에 의한 한국산 며느리밥풀속(Melampyrum L.)의 분류학적 재검토

요약 : 한국산 며느리밥풀속(Melampyrum L.) 6분류군(M. roseum, M. roseum var. japonicum, M. roseum var. ovalifolium, M. setaceum, M. setaceum var. nakaianum, and M. koreanum) 을 대상으로 외부형태학적 형질을 관찰하였다. 조사 결과, 잎의형태, 포의 형태, 포 가장자리 강모의 분포 위치와 수는 유용한검색형질로 확인되었으며, 줄기의 횡단면 형태와 털의 분포, 포의 기부 형태, 꽃받침 열편과 정단부 돌기의 형태, 화관의 색과길이는 유용한 검색형질로 나타났다. 이러한 외부형태학적 형질을 바탕으로 한국산 며느리밥풀속의 검색표를 제시하였다.

Dormancy-breaking and Germination of Chelidonium majus L. subsp. asiaticum H. Hara Seeds

Abstract: The demand for Chelidonium majus L. subsp. asiaticum H. Hara is expected to increase due to its pharmacological properties such as antibacterial, antioxidant, and anti-inflammatory effects. However, an effective propagation system for this species has not yet been established. This study was conducted to analyze the seed dormancy and germination characteristics of C. majus L. subsp. asiaticum H. Hara native to Korea and establish a mass propagation system. The dormancy type was primarily classified by analyzing the general information of the collected seeds. The seed dormancy breaking was investigated by comparing the effects of cold stratification (0, 2, 4, 8, 10, or 12 weeks) with warm stratification (S, summer temperature, 25/15°C) and intermediate temperature stratification (A, autumn temperature, 15/10°C) of alternating temperature stratification (S12-A4 or S12-A8 weeks). After dormancy break, 500 ㎎/L GA3 and GA4+7 treatment replaced cold stratification and improved seed germination. The results of this study are expected to provide basic data for future seed propagation and mass propagation by analyzing the dormancy and germination characteristics of C. majus L. subsp. asiaticum H. Hara seeds.

쉬나무의 용기묘 대량생산을 위한 적정 시비 수준 구명

초록 본 연구는 대체에너지, 생태복원 및 밀원식물용 등으로 이용되어 묘목 수요가 증가되고 있는 쉬나무의 우량한 용기묘 생산을 위한 적정 시비수준을 구명하고자 실시하였다. 공시용기는 임업시설양묘용 플라스틱 용기(350 ㎖/구)를 사용하였다. 시비처리는 무시비구와 함께 수용성 복합비료인 Multifed 19 (N:P:K=19:19:19, v/v)를 50 ㎎·L -1 , 100 ㎎·L -1 , 150 ㎎·L -1 , 200 ㎎·L -1 로 조절하여 시비하였다. 실험결과, 간장과 근원경 생장은 시비수준이 높아짐에 따라 높은 생장값을 나타내다가 100 ㎎·L -1 이상의 시비에서는 낮아지는 경향을 보였다. 뿌리발달은 100 ㎎·L -1 에서 가장 왕성하였다. 건물생산량도 100 ㎎·L -1 에서 가장 높았고 전체적으로 간장 및 근원경 생장 결과와 유사한 경향으로 나타났다. 묘목의 품질을 나타내는 지수인 QI (Quality Index)도 100 ㎎·L -1 에서 0.97로 다른 처리구 보다 높게 조사되었다. 연구결과를 종합하면, 쉬나무 용기묘의 우량한 묘목생산을 위한 적정 시비수준은 100 ㎎·L -1 로 판단되며, 쉬나무 용기묘의 대량생산을 위한 기초자료로 활용될 것으로 기대된다

효율적인 도라지 추출물 제조를 위한 복합 효소 처리 방법 연구

Abstract: The two glycoside hydrolases (Viscozyme and Termamyl 2X, commercial food grade enzymes) was used for production of balloon flower (Platycodon grandiflorum) extract. The complex enzyme was prepared in a ration of Viscozyme and Termamyl 2X of 2:1. The optimal temperature and concentration of complex enzyme was 50o C and 5% (w/w). Under optimal conditions, the extraction yield, crude saponine, and platycodin D contents of complex enzyme treated Platycodon grandiflorum extract was 43, 24, and 303% higher than Platycodon grandiflorum hot water extract. And the DPPH radical scavenging activity, ABTS scavenging activity, tyrosinase inhibitory activity, and elastase inhibitory activity of the complex enzyme treated Platycodon grandiflorum extract exhibited 1.7, 1.3, 1.4, and 1.4 fold higher than Platycodon grandiflorum hot water extract. These results indicated that the complex enzyme treatment can be used as an economical method for industrial production Platycodon grandiflorum extract. And the complex enzyme treated Platycodon grandiflorum extract cad potentially be used for developing a new cosmetic or health food ingredients.

Effect of experimental climate change and soil moisture content on germination and early growth

ABSTRACT: This study was conducted to investigate the effect of changes in soil moisture content under climate change conditions on the growth response of Quercus dentata, a representative species of the genus Quercus on the Korean peninsula. The results showed that Q. dentata germination increased by 9.4 days under climate change conditions (RCP 4.5 scenario), whereas the below-ground biomass and plant biomass increased by 1.3 and 1.2 times, respectively. However, the soil moisture treatment condition solely affected the leaf area, below-ground biomass, and plant biomass but not the germination percentage, germination speed, aboveground length, and above-ground biomass. The variance across the measured growth variables was considerably higher in the control group than in the climate change treatment group, whereas the variance was lower for lower soil moisture content. The findings suggested that under climate change conditions, Q. dentata germinated rapidly and exhibited broader leaf area and increased development below ground, which may be advantageous for the early establishment of seedlings. However, the plants may face difficulties in conditions with low soil moisture content. In conclusion, with continuous climate changes, Q. dentata seedlings are predicted to efficiently adapt to the altered soil moisture content to exhibit even more reliable growth than with the current condition. However, the growth of Q. dentata seedlings would be more difficult with severely low soil moisture content than with abundant soil moisture content, which is predicted to be detrimental to population maintenance in the long term.

히어리 자생지 식생구조와 환경요인 간 상호관계

Abstract : The purpose of this study is to understand the vegetation structure of the Corylopsis coreana habitats in South Korea and their correlation with environmental factors, in order to provide basic data for preparing conservation. A total of 40 vegetation survey plots were established in around Southern region, Gangwon-do and Gyeonggi-do Province. The cluster analysis revealed four distinct clusters: Quercus mongolica-Pinus densiflora (QmPd), Castanea crenata (Cc), Quercus mongolica (Qm), and Pinus koraiensis (Pk). Among them, the QmPd cluster was selected as the representative community. Soil texture analysis that most areas consisted of loamy soil and were distributed on the northern aspects. The plantation, represented by Pk, exhibited a high content of exchangeable aluminum (5.227±0.342 ㎎/㎏), suggesting the need for soil improvement and monitoring in these habitats. The canopy openness (forest gap) ranged from 11% to 21%, indicating a relatively closed canopy in many survey plots. Non-metric multidimensional scaling analysis indicated heterogeneous species composition between the QmPd cluster and the Pk cluster (total R2 = 0.608).

COLD STRATIFICATION BREAKS THALICTRUM UCHIYAMAE NAKAI SEED DORMANCY VIA PROTEOMIC CHANGES

ABSTRACT This study was conducted to obtain basic data related to the seeds, germination conditions, and physiological changes in the seeds of Thalictrum uchiyamae Nakai (Seoul Meadow Rue). The seed was elongated and tapering at both ends, and its length and width were 1.810–2.744 and 0.576–0.989 mm, respectively. When the seeded seeds were planted immediately, the germination rate was less than 5% at 15/6°C or 20/10°C, but after cold stratification for 8 weeks at 5°C, the germination rate exceeded 80% at 30/20°C, 20/10°C, and 15/6°C. Thus, it was ed that T. uchiyamae seeds are morphophysiologically dormant. The physiological changes in the seeds during cold stratification were investigated in terms of proteomics; the number of activated proteins increased from 330 to 353 after cold stratification. We ed that the disappearance of or decrease in tunicamycin and phytochrome is associated with the breaking of dormancy in T. uchiyamae seeds.
