Species-Specific seed vigor test of aging chive for restoration and regional adaptation under climat

ABSTRACT This study aimed to develop a vigor test method that can be applied in the seed storage and ecological restoration of Aging chive (Allium senescens L). The impact weight of each temperature on germination was described with a sine curve equation. The closer the temperature was to 18.9 °C, the faster the germination was, and temperatures below 8.0 °C and above 29.7 °C negated the temperature accumulation effect. A verification and validation test ed the 'impact weight on germination' to be reasonable because it exhibited a hump-shaped relationship with the time to reach 85% germination among the temperature treatments. Furthermore, the time to reach 25, 50 and 75% germination and the change in germination with time did not differ between the actual and predicted values. The hourly accumulated value of 'impact weight on germination' was taken as the temperature score (TS), and the temperature requirement for vigorous germination was 294TS. The temperature requirement was applied to hourly temperature data from Geumsan, Korea, from 1999-2018, and the average date of vigorous seed germination was May 3. Increasing temperature elevation advanced the predicted germination date. Based on the earliest predicted date, germination will advance rapidly. If the temperature is elevated by 2.5 °C, the germination date will advance markedly to November 23.

싸리나무 추출물의 Xanthoangelol 분리 및 HPLC-DAD 밸리데이션

Abstract This study was undertaken to determine the characteristics of xanthoangelol, the major chalcone constituent derived from the extracts of different parts of Lespedeza bicolor . Xanthoangelol was isolated from the root extract using column chromatography and used as a standard for quantitative analysis. The structure of the isolated compound was established based on spectroscopic evidence. The HPLC-DAD method was validated for specificity, linearity, precision, accuracy, limit of detection, and limit of quantitation. The calibration curve of xanthoangelol had significant linearity (R 2 >0.9999). Limit of detection and limit of quantitation 0.018 and 0.059 µg/mL, respectively. The relative standard deviation values of precision test, and intra- and inter-day tests were less than 0.22 and 0.40%, respectively. In the recovery test, the accuracy ranged from 98.98-102.78% with RSD values less than 0.13%. The method validation parameters indicate the applicability of the HPLC method for quality control of food or drug formulations containing L. bicolor.

Physiological Activity of Methanol Extracts from Sambucus sieboldiana var.

ABSTRACT Sambucus sieboldiana var. miquelii (Nakai) Hara is distributed in Korea, China, and Japan, and has been used as an anti-rheumatic in folk medicine in oriental countries. The present study aims to investigate the potential use of this species in health functional foods, cosmetics, and food preservatives. Methanol extracts of leaves and branches from this plant were prepared to quantitatively analyze the total phenol and flavonoid contents, and to investigate the antioxidative and enzyme inhibitory activities, and the inhibition of nitric oxide (NO) production activity. The results showed that the total polyphenol and flavonoid contents of the crude extract were 1.52±0.1 mg/g and 1.73±0.1 mg/g, respectively. S. sieboldiana polyphenols exhibited potent scavenging activity shown by 2, 2'-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay and 2, 2-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical cation assay. The crude extract also exhibited significant α-glucosidase and tyrosinase inhibitory activity with IC 50 values of 183.5 μg/ml and 323.9 μg/ ml, respectively. Additionally, the crude extract exhibited strong anti-inflammatory activity determined through the nitric oxide inhibition assay in a dose-dependent manner with an IC 50 value of 36.7 μg/ml and no cytotoxic effect on the macrophages. Therefore, we demonstrated that the leaves and branches of S. sieboldiana extract possess antioxidant, anti-diabetic, depigmentation potential, and NO production inhibitory activities. According to recent research, S. sieboldiana has great potential as a source of the bioactive compound which could be used as food, cosmetics, and pharmaceutical agents.

Anti-aging and Anti-inflammatory Activities of the Extracts of Calamagrostis arundinacea

ABSTRACT The anti-aging and anti-inflammatory activities of hot-water (Ca-HW) and 70% ethanol (Ca-E70) wholeplant Calamagrostis arundinacea extracts, as well as their bioactive potentials, were investigated using cell-free and cell-mediated experimental systems. Use of the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical decolorization assay to evaluate the antioxidant activity of the Ca-HW and Ca-E70 extracts revealed DPPH radical scavenging activities of 27% and 48%, respectively. Neither extract caused significant cytotoxicity, and both showed cell proliferation and promotion effects using RAW 264.7, B16F10, and CCD986sk cells. B16F10 melanoma cells showed higher melanin synthesis when treated with 100 mg/ml Ca-HW or Ca-E70 than with arbutin, indicating a stronger inhibitory effect of arbutin on melanin synthesis. Ca-HW and Ca-E70 increased pro-collagen biosynthesis in the human fibroblast CCD986-SK cell line by 24.69% and 12.55%, respectively. Analysis of the anti-inflammatory effects of different concentrations of Ca-HW and Ca-E70 in RAW264.7 cells revealed that Ca-E70 appeared to inhibit the lipopolysaccharide-induced production of nitric oxide and IL-6, a proinflammatory cytokine; therefore, Ca-E70 showed an anti-inflammatory effect. These results suggested that C. arundinacea extracts could have skin anti-aging and anti-inflammatory properties.

Improved Biotransformation of Platycoside E into Deapiose-Xylosylated Platycodin D by Cytolase PCL5

ABSTRACT Platycosides are the functional saponins present in balloon flowers that exert diverse biological effects, and which can be further improved by their deglycosylation. Deapiose-xylosylated platycodin D, which is absent in balloon flowers, can be generated only by cytolase PCL5 by acting on platycoside E. To improve cytolase PCL5-catalyzed production of deapiose-xylosylated platycodin D from platycoside E, we explored the use of high hydrostatic pressure (HHP). At an HHP of 150 MPa, the optimal temperature of cytolase PCL5 activity for converting platycoside E into deapiosexylosylated platycodin D shifted from 50 to 55 C, and increased the activity and stability of the enzyme by 5- and 4.9-fold, respectively. Under HHP, the enzyme completely converted 1 mM platycoside E into deapiose-xylosylated platycodin D within 4 h, with a 3.75-fold higher productivity than that under atmospheric pressure. Our results suggest that the application of HHP is a potential method for the economical production of platycosides and enzyme-catalyzed biotransformation of functional saponins.

Does oxygen affect ageing mechanisms of Pinus densiflora seeds?

Abstract: During desiccation, the cytoplasm of orthodox seeds solidifies into an intracellular glass with highly restricted diffusion and molecular mobility. Temperature and water content govern seed ageing rates, while oxygen (O2) can promote deteriorative reactions. However, whether the cytoplasmic physical state affects involvement of O2 in seed ageing remains unresolved. We aged Pinus densiflora seeds by controlled deterioration (CD) at 45 °C and distinct relative humidity (RH), resulting in cells with a glassy (11% and 30% RH) or fluid (60% and 80% RH) cytoplasm. Hypoxic conditions (0.4% O2) during CD delayed seed deterioration, lipid peroxidation, and decline of antioxidants (glutathione, α-tocopherol, and γ-tocopherol), but only when the cytoplasm was glassy. In contrast, when the cytoplasm was fluid, seeds deteriorated at the same rate regardless of O2 availability, while being associated with limited lipid peroxidation, detoxification of lipid peroxide products, substantial loss of glutathione, and resumption of glutathione synthesis. Changes in metabolite profiles provided evidence of other O2-independent enzymatic reactions in a fluid cytoplasm, including aldo-keto reductase and glutamate decarboxylase activities. Biochemical profiles of seeds stored under seed bank conditions resembled those obtained after CD regimes that maintained a glassy cytoplasm.
