New diploid populations of the Chrysanthemum zawadskii complex (Asteraceae) in natural habitats of Korea

Absract : Chrysanthemum zawadskii Herbich is widely distributed from northern Europe to East Asia. It has been reported that various ploidy levels ranging from diploid (2n = 2x = 18) to decaploid (2n = 10x = 90) are distributed in Korea. The diploid populations of C. zawadskii, however, have limited habitats, with only four populations from two locations compared to polyploids that are widely distributed in Korea. During recent biodiversity research on the C. zawadskii complex, new habitats of diploid populations of C. zawadskii not reported earlier in Korea were identified. To understand the speciation mechanism accompanying polyploidization of the C. zawadskii complex, it was necessary to investigate the features of these new populations as to whether they identical to or different from previously reported diploid populations. For this purpose, we identified the number and karyotypes of somatic chromosomes in four new diploid populations and compared the morphological features. It was found that the new diploid populations differed slightly in terms of the size and color of the flowers and in the shape of the leaves, with continuous variations. Representative karyotypes of the new populations were included in previously reported diploid karyotypes, but the numbers and locations of satellite chromosomes differed. In addition, we found that one of them is a sympatric population with different ploidy levels, including diploid, tetraploid, and hexaploid. From these results, it was confirmed that morphological and cytological variations exist among diploid populations in the C. zawadskii complex.

우산나물(Syneilesis Palmata Maxim) 종자 추출물의 항산화 및 α-glucosidase 저해 활성

요약 : 우산나물은 한반도 전역에서 자라 과거에서부터 산나물 및 민간의약품으로 활용된 식물이다. 본 연구는 기존에 연구되지 않은 Syneilesis palmata Maxim 종자를 대상으로 항산화 활성 및 alpha-glucosidase 저해 활성을 분석하였다. 추출물은 2018년에 경상북도 봉화에서 수집된 종자로부터 준비되었다. 우산나물 종자 추출물 내 총 페놀성 화합물의 함량은 MeOH 추출물에서 11.23 mg GAE/g seeds, 총 플라보노이드 함량은 MeOH 추출물에서 3.69 mg QE/g seeds로 가장 높게 나타났다. 우산나물 종자 추출물의 DPPH 라디컬 소거능의 EC50은 MeOH 추출물에서 67.0 μg/mL으로 가장 높게 나타났으며, ABTS 라디컬 소거능의 EC50은 24.7 μg/mL로 MeOH 추출물에서 가장 높게 나타났다. 우산나물 종자 추출물의 FRAP value는 1354.5 μM Fe(II)/g extract으로 MeOH 추출물에서 가장 높게 나타났다. 우산나물 종자 추출물의 α-glucosidase 저해활성 IC50은 MeOH 추출물에서 17.43 μg/mL로 가장 높게 나타났다. 또한, 높은 α-glucosidase 저해활성을 가진 추출물에서 개별 페놀성 화합물들이 검출되었다. 본 연구결과를 통해 풍부한 페놀성 물질을 가진 우산나물 종자의 MeOH 추출물이 항산화와 항당뇨 효과를 나타낸다는 것을 확인하였으며, 이는 효과적인 건강기능식품 소재 및 천연 의약품으로서 이용할 수 있을 것으로 기대한다.

A Study on the Genetic Variations and Germination Characteristics of Rhododendron sohayakiense to Prepare for Climate Change Threat

Abstract : Rhododendron sohayakiense is an endemic and near-threatened species (Korean Red List, NT) found in the alpine regions of South Korea that requires conservation. This study investigated the species' genetic variations and seed germination characteristics and predicted its potential habitat change according to climate change scenarios. The genetic diversity of R. sohayakiense at the species level (P = 88.6%; S.I. = 0.435; h = 0.282) was somewhat similar to that observed for the same genus. The inter-population genetic differentiation was 19% and revealed a relatively stable level of gene exchange at 1.22 in each population. The main cause of gene flow and genetic differentiation was presumed to be the Apis mellifera pollinator. Seed germination characteristics indicated non-deep physiological dormancy, with germination at ≥10 °C and the highest percent germination (PG) of ≥60% at 15–25 °C, while the PG was ≥50% at 30 °C. The PG increased at constant temperature than at variable temperatures; the mean germination time decreased as temperature increased. The climate scenarios SSP3 and SSP5 were analyzed to predict future R. sohayakiense habitat changes. The variables of the main effects were Identified as follows: elevation > temperature seasonality > mean diurnal range.

Effect of Propagation Media and Plant Growth Regulators on Adventitious Rooting of Softwood Cuttings of Apocynum lancifolium Russanov

Abstract : Background and objective: Apocynum lancifolium Russanov is a perennial herb native to Korea and belonging to the familyApocynaceae. Herein, cutting mediums and plant growth regulators (PGRs) were examined to investigate the requirementsfor the cutting propagation of A. lancifolium. Methods: The study was conducted over a period of 6 weeks from May to June 2023. As types of cutting medium forpropagation by cutting, Kanuma soil, commercial potting mixes, perlite, and peat moss were used, and the PGRs ofindole-3-butyric acid (IBA) at varying concentrations (100, 500, and 1,000 mg⋅L-1) and a 1:1 (v/v) solution of Rootone(0.4% 1-naphthylacetamide) diluted in water were applied for this study. After six weeks, growth characteristics, includingcutting survival rate, rooting rate, shoot height, the number of leaves, root length, the number of roots, dry weight ofshoots, and dry weight of roots, were examined. Results: Among the different growing substrates used for experimentation, the cutting survival rate on Kanuma soil (81.7 ±5.0%) was the highest, followed by commercial potting mixes (40.0 ± 7.2%) and perlite (66.7 ± 2.7%), while the lowestcutting survival rate was on peat moss (35.0 ± 6.3%). On treatment with Kanuma soil (81.7 ± 5.0%), the rooting rate wasalso significantly greater than that when treated with other substrates. In the PGR experiment, the cutting survival rate androoting rate treated with Rootone (0.4% 1-naphthylacetamide) were significantly higher than those when treated withother substrates. Further, the number of roots increased with an increase in the IBA concentration. Based on the results,we recommend treating A. lancifolium cuttings with Rootone and Kanuma soil to ensure successful cutting propagation ofA. lancifolium. Conclusion: This mass production system for A. lancifolium via cutting propagation shows promise in terms of scalabilityand could help with germplasm conservation efforts for A. lancifolium.

반려식물 키트 산업의 활성화 방안에 관한 연구 - 산업 종사자의 수요를 중심으로 -

요약 : 본 연구의 목적은 반려식물 키트 산업의 현황을 파악하고, 산업 활성화를 위한 지원 정책 우선순위를 알아보는 것이다. SWOT 분석을 통해 반려식물 키트 산업의 현황을 진단하였고, 산업 활성화를 위한 지원 정책 우선순위 파악을 위해 반려식물 키트 산업 종사자를 대상으로 계층분석을 활용하였다. SWOT 분석 결과, SO 전략으로는 정부의 지원 정책을 활용하여 마케팅을 강화하고 친환경 이미지와 DIY 키트를 결합한 제품을 개발하는 것이며, WO 전략으로는 시장 인지도를 높이기 위한 광고 캠페인을 전개하고 유통망 확장을 위한 전략적 파트너십을 구축하는 것으로 나타났다. 또한, ST 전략으로는 경제 불확실성에 대응하여 가격 경쟁력을 강화하고 경쟁 제품 대비 차별화된 고유 가치를 제안하는 것이며, WT 전략으로는 비용 절감을 위한 생산 공정을 개선하고 소비자 피드백을 반영하여 제품 품질을 개선하는 것으로 나타났다. AHP 분석 결과 총 3개의 상위항목과 12개의 하위항목으로 구성한 평가 항목을 도출하였고, 전문가 설문을 통해 총 17부의 데이터를 수집하였다. 분석 결과, '진입 단계' (0.482), '활성화 단계' (0.397), '고도화 단계' (0.121) 순의 중요도가 도출되었다. 또한 '간담회 개최'(0.181)가 가장 중요한 하위항목으로 나타났으며, '키트 개발 지원'(0.020)은 상대적으로 낮은 중요도를 보였다. 반려식물 키트 산업은 정원 산업 발전을 위한 초기 단계 사업으로 적절한 정책적 인큐베이팅은 정원 산업 활성화에 도움을 줄 수 있다. 본 연구의 결과는 반려식물 키트 산업 활성화를 위한 산업계의 수요에 대한 기초 정보를 제공할 수 있다.

Production of Daidzein and Genistein from Seed and Root Extracts of Korean Wild Soybean

Abstract: Isoflavone glycosides are commonly biotransformed into isoflavone aglycones due to the superior biological activities of the latter. Wild soybeans contain a higher isoflavone content than domesticated soybeans due to their high level of genetic diversity. In this study, we cloned and characterized a thermostable β-galactosidase from the extreme thermophile Thermoproteus uzoniensis for potential application in isoflavone conversion in Korean wild soybeans. The purified recombinant enzyme exhibited a maximum specific activity of 1103 µmol/min/mg at pH 5.0 and 90 ◦C with a half-life of 46 h and exists as a homodimer of 113 kDa. The enzyme exhibited the highest activity for p-nitrophenyl (pNP)-β-D-galactopyranoside among aryl glycosides and it hydrolyzed isoflavone glycosides in the order genistin > daidzin > ononin > glycitin. The enzyme completely hydrolyzed 2.77 mM daidzin and 3.85 mM genistin in the seed extract of wild soybean after 80 and 70 min with productivities of 1.86 and 3.30 mM/h, respectively, and 9.89 mM daidzin and 1.67 mM genistin in the root extract after 180 and 30 min, with the highest productivities of 3.30 and 3.36 mM/h, respectively, compared to other glycosidases. Our results will contribute to the industrial production of isoflavone aglycone using wild soybean and this is the first report on the enzymatic production of isoflavone aglycones from isoflavone glycosides in wild soybeans.

Non-deep physiological dormancy in seeds of Euphorbia jolkinii Boiss. native to Korea

Abstract Background: Euphorbia jolkinii Boiss. is a perennial species native to Jeju Island and the southern coastal area of Korea. Particularly on Jeju Island, the yellow flowers of E. jolkinii Boiss. have a high ornamental value because of their contrast with basalt. This study was conducted to investigate the effects of different temperatures (5, 15, 20, and 25 °C) and gibberellic acid (GA3) concentrations (0, 10, 100, or 1000 mg/L) on seed dormancy and germination of E. jolkinii. In addition, we classified the seed dormancy type and compared types with those of other species in the same genus. Results: The number of seeds with viable embryos and endosperms was approximately 66%. The final germination percentages at 5, 15, 20, and 25 °C were 51.7%, 83.5%, 2.6%, and 0.0%, respectively. In GA3 concentration experiments, the final germination percentages of 0, 10, 100, and 1000 mg/L were 83.5%, 91.7%, 79.1%, and 83.4%, respectively, at 15 °C conditions, and 0.0%, 6.9%, 13.2%, and 27.3%, respectively, at 25 °C. Conclusions: Germination improved at temperatures of 15 °C or lower. Furthermore, GA3 treatment effectively reduced germination times. Thus, the seeds of E. jolkinni were classified as having non-deep physiological dormancy.

Effects of culture medium composition on the in vitro growth of stem cuttings of Kirengeshoma korean

Background and objective: Kirengeshoma koreana Nakai is an endemic and endangered species in South Korea. We conducted in vitro propagation and regeneration of K. koreana from stem cuttings to investigate the effects of nine different basal culture media and five different carbohydrate sources on its growth. Methods: Apical segments (at least 1 cm long) collected randomly from a six-week-old K. koreana plantlet grown in vitro were used as explants. In the first experiment, the explants were transferred into square vessels containing 50 mL of nine different basal culture media supplemented with 30 g⋅L-1 sucrose and 3 g⋅L-1 Phytagel. In the second experiment, the explants were transferred into square vessels containing 50 mL half-strength SH medium supplemented with five different carbohydrate sources at 30 g⋅L-1. Each medium was solidified with 3 g⋅L-1 Phytagel. All experiments contained 4 cultures, and the experiments were repeated four times to enhance reproducibility. Data on stem length, shoot fresh weight, leaf width, leaf length, root count, and root length were collected at the end of 8 weeks of culture. Results: ANOVA showed that the basal culture medium had a significant effect on K. koreana growth (p half-strength SH medium was the best condition for stem length, shoot fresh weight, leaf width and root length (3.76 ± 0.12 cm, 0.60 ± 0.06 g, 1.19 ± 0.05 cm, and 2.83 ± 0.13 cm, respectively). However, the highest percentage increase in root count (13.00 ± 0.90) was found to occur with half-strength WPM. The effect of different carbohydrate sources on K. koreana growth was significantly different (p .09, respectively). Maltose was the best condition for shoot fresh weight (0.90 ± 0.09 g). Although there was no significant difference, sucrose was found to be best for leaf width, leaf length and root length (1.34 ± 0.07 cm, 2.34 ± 0.10 cm, and 3.86 ± 0.19 cm, respectively). Conclusion: This in vitro propagation and regeneration system for K. koreana shows promise in terms of scalability and could help greatly with germplasm conservation and restoration efforts for the species.

남산 소나무군락의 토양호흡과 환경요인인 기온과 강수량과의 상호관계 및 도시림 관리의 의미

요약 본 연구에서는 대도시공원 내 소나무군락의 토양권의 유기탄소 배출량을 정량화하여 기후요인과의 관계를 분석하고 이를 통해 토양호흡량 배출을 감축하기 위한 관리방안을 마련하고자 하였다. 이를 위해 서울시 남산에 위치하고 있는 소나무 군락의 토양호흡량의 유기탄소량을 조사하고 환경요인과 연결시켜 관련성을 알아보았다. 그 결과 남산 소나무군락의 토양 호흡량의 평균 유기탄소량은 7.978 ton C ha -1 yr -1 로 2011년에 6.893 ton C ha -1 yr -1 에서 2018년에 8.660 ton C ha -1 yr -1 로약 0.682 ton C ha -1 yr -1 만큼 증가하였다. 또한, 봄철(3월-5월)의 강수량은 토양호흡의 증감을 설명하는 환경요인 중 하나였다. 이는 도시림의 생태적 관리를 위해서는 토양함수량이 높아지지 않도록 배수관리가 중요함을 시사한다.
