Cloning and characterization of α-L-rhamnosidase from Chloroflexus aurantiacus and its application

Abstract Objective This study was conducted to characterize recombinant a-L-rhamnosidase from Chloroflexus aurantiacus and apply the enzyme in the production of isoquercitrin from rutin. Results The a-L-rhamnosidase from C. aurantiacus was cloned and expressed in Escherichia coli BL21 and purified as a soluble enzyme. a-L-rhamnosidase purified from C. aurantiacus has a molecular mass of approximately 105 kDa and is predicted to exist as a homodimer with a native enzyme of 200 kDa. The purified enzyme exhibited the highest specific activity for rutin among the reported isoquercitrin producing a-L-rhamnosidases and was applied in the production of isoquercitrin from rutin. Under the optimised conditions of pH 6.0, 50 C, 0.6 U mL-1 a-L-rhamnosidase, and 30 mM rutin, a-L-rhamnosidase from C. aurantiacus produced 30 mM isoquercitrin after 2 h with a 100% conversion yield and productivity of 15 mM h-1. Conclusions We achieved a high productivity of isoquercitrin from rutin. Moreover, these results suggest that a-L-rhamnosidase from C. aurantiacus is an effective isoquercitrin producer.

A new lichen-forming fungus, Orientophila corticola, from South Korea, with a key to the genus

Abstract Orientophila corticola is described as a new lichen-forming fungus from South Korea. The new species is distinguishable from other Orientophila species by the substrate preference to tree barks and the habitat specificity to inland areas. Molecular analyses applying internal transcribed spacer (ITS) and mito-chondrial small subunit(mtSSU) sequences clearly classify O. corticola as a distinct species in the genus Orientophila. A surrogate key is provided to assist in the identification of all 13 taxa in the genus Orientophila.

Arthonia dokdoensis and Rufoplaca toktoana - Two New Taxa from Dokdo Islands(South Korea)

Abstract Arthonia dokdoensis sp. nov., a lichenicolous fungus from the subcosmopolitan Arthonia molendoi complex growing on crustose thalli of species of the genus Orientophila (subfamily Xanthorioideae, Teloschistaceae), as well as the lichen species Rufoplaca toktoana sp. nov.(subfamily Caloplacoideae, Teloschistaceae) similar to Rufoplaca kaernefeltiana , both from Dokdo Islands, Republic of Korea, are described, illustrated, and compared with closely related taxa. In the phylogenetic tree of the Arthoniaceae based on 12S mtSSU and RPB2 gene sequences, the phylogenetic position of the A. dokdoensis and the relationship with the A. molendoi group are illustrated, while the position of the newly described R. toktoana is ed by phylogenetic tree based on ITS nrDNA data.

The complete chloroplast genome of hemi-parasitic Pedicularis hallaisanensis (Orobanchaceae)

ABSTRACT We determined the complete chloroplast genome of Pedicularis hallaisanensis (Orobanchaceae), a hemiparasitic perennial herb. This genome is 143,469 bp long and features a large single-copy region(81,664 bp) and a small single-copy region (12,203 bp), separated by two inverted-repeat regions(24,801 bp each). It contains 115 genes – 70 for coding, eight for rRNA, and 37 for tRNA. However, 11 ndh genes have been pseudogenized, truncated, or deleted. Our phylogenetic tree showed that these hemi-parasitic plants are sister to holo-parasitic genera within Orobanchaceae.

The First Report of Fusarium solani Causing Wilting in Cnidium officinale in Korea

Cnidium officinale is a perennial herb belonging to the family Umbelliferae. Its origin is China and is also distributed in Korea and Japan. In 2017, a phenomenon of browning and wilting of leaves was discovered in the cultivation field of the National Institute of Forest Science, Suwon, Korea. The pathogens isolated from plants were consistent with Fusarium solani as a result of morphological examination and molecular phylogenetic analysis. This is the first record of F. solani on C. officinale in Korea.

중요생물다양성지역(KBAs) 기준 적용을 통한 설악산 보호구역 평가

Abstract This study was aimed to design core areas applied by the global conservation criteria to promote the public awareness to the protected areas and the value cognition of the Key Biodiversity Areas (KBAs), targeting the Mt Seorak, according to the designation of globally important biodiversity areas. As a method for carrying out this study, the biota were cataloged through literature reviews and field trips. With applied by the Global Red List criteria of the International Union for Conservation of Nature (IUCN), only nine species were categorized in the studied area; plants were classified into six species as follows: Megaleranthis saniculifolia ohwi, Bupleurum euphorbioides Nakai, Hanabusaya asiatica Nakai, Thuja koraiensis Nakai, Leontopodium leiolepis Nakai, Androsace cortusaefolia Nakai, fish was classified one species as follow: Pungitius sinensis Tanaka, and the mammal was classified as two species as follows: Hydropotes inermis, Naemorhedus caudatus. According to the occupation area (EoO, Extent of Occurrence) and Minimum Viable Population(MVP), the size of protected area was 234.56㎢ for plants, 235.07㎢ for mammals, and 0.14㎢ for fish, and the Key Biodiversity Area(KBA) of Mt. Seolak suggested as 286.72㎢.
